Our biorepository space at The Anlyan Center (TAC) is one block from Yale-New Haven Hospital. It has 24-hour accessibility by key card. TAC houses many prominent basic scientists within Yale University School of Medicine. It is a state-of-the-art building complex completed in 2001 with six floors of laboratories for disease-oriented research, teaching facilities, and core research resources.
We have over 30 feet of dedicated bench space for specimen processing and biomarker assays. At present, the repository consists of nine Thermo Revco Ultima Plus -86C freezers with the physical and electrical capacity for at least four more units. Of the nine freezers, three, containing archived biosamples, are stored in our freezer farm and the remaining six, in the laboratory. Each unit is plugged into the emergency backup AC power with less than a one-minute response time if standard power failure occurs.
The units are additionally connected to an analog Sensaphone 400 remote monitoring system (with battery backup) that is programmed to call a phone tree of four staff members if any of the freezers' preset temperature is exceeded or if power failure occurs. All freezers in the repository receive biannual preventive maintenance, and the system is tested on a quarterly basis to ensure proper operation. The repository contains biospecimens that have been (and continue to be) collected in the course of eight studies. It holds approximately half a million biospecimen aliquots for more than 4,000 patients from several multicenter studies and includes the following source materials: blood, urine, DNA, and perfusate collected during the course of various kidney disease studies; there are no restrictions on the nature of the materials being collected.

Biomarker Research Laboratory
The laboratory has a fully automated clinical chemistry analyzer, Rx Daytona from Randox Laboratories (Ireland, UK) which is used to measure electrolytes, cystatin C, microalbumin, albumin, urea, creatinine, and bicarbonate. The laboratory is also equipped with a MESO QuickPlex SQ 120 instrument from MSD and a Synergy HT multi-mode microplate ELISA reader from BioTek Instruments. Other supporting equipment include a Sorvall legend bench top centrifuge, microplate shakers (Corning and Fisher Scientific), a shaker incubator (The Jitterbug from Boekel), refrigerators and -20 C freezers for processing and storing samples and reagents. All the instruments are connected to individual desktops with inbuilt software for the recording and analysis of data. Additionally, the biomarker laboratory works closely with the other central facilities, such as the Yale West Campus Analytical Core facility for measuring various diuretics in urine and blood samples, using the Q-TOF mass spectrometer. The Yale Metal Geochemistry Center at The Department of Geology & Geophysics provides measurements of endogenous lithium in blood and urine by ICP-MS.
The PATR laboratory runs a wide range of multiplex biomarker assays using the Meso Scale Platform (MSD). Some of the assays that are currently being offered on the MSD platform are:
- 7-plex kidney injury panel -5 (UMOD, NGAL, Albumin, Cystatin C, B2M, EGF, OPN)
- 4-plex (IL-18, MCP-1, KIM-1 and YKL-40)
- 2-plex TGF R1 and TGF R2
- 1-plex NGAL
